چکیده
In the past 3 years techniques have been developed for autoradiographic localization of diffusable compounds including neurotransmitter receptor ligands (Young and Kuhar, 1979; Herkenham and Pert, 1980). The high spatial resolution of these techniques has provided elegant demonstrations of the regional heterogeneity of receptor densities within the CNS, however, this is achieved at the expense of the direct quantitation capabilities of homogenate binding methods. We have, therefore, developed an autoradiographic technique using tritium sensitive film (LKB Ultrofilm 3H) to achieve rapid high resolution quantification of benzodiazepine, GABA and glycine receptor numbers and affinities in multiple areas of the central nervous system. For the receptor assays, frozen sections of brain and spinal cord were mounted on gelatin coated slides at room temperature and stored at -20°C. Prior to assay, the slides were given three consecutive 5 min washes in 50 mM Tris-citrate buffer (pH 7.2) at 0°C. They were then incubated for 30 min at 0°C in solutions of either [3H]flunitrazepain (1-15 nM, 88 Ci/mmol) or [3H]muscimol (5-80 nM, 19 Ci/mmol) in 50 mM Tris citrate buffer pH 7.2, or [3H]strychnine (5-80 nM, 15 Ci/mmol) in 50 mM Na-K phosphate buffer (pH 7.0). Adjacent sections were incubated with excess diazepam (2>(10 -6 M), GABA (10 -3 M) or

Quantitative a U T O ~ D I O G ~ H Y of L-i3higlutamate Binding to Rat Brain
A technique has been developed to investigate sodium-independent t-lJHJglutamate binding in rat brain sections using quantitative autoradiography and tritium-sensitive film. Binding is rapid (reaching equilibrium in 5 rain) and reversible (having a 11/2 of dissociation of 0.38 rain). Glutamate apparently bound to a single population of sites with a Kd of about 1.0 ~M. The pharmacology of this b...

A quantitative autoradiographic technique for measuring the binding of [3H]muscimol to central nervous system GABA receptors is described using tritium-sensitive film. [3H]Muscimol binding was studied in primary and secondary striatal projection areas of rat brain following kainic acid lesions of the striatum. Seven days after the lesion, binding affinities in the striatum and its projection ar...
متن کاملQuantitative autoradiography of [3H]sulpiride binding sites in rat brain.
A technique has been developed to investigate [3H]sulpiride binding in rat brain sections using quantitative autoradiography and tritium-sensitive film. Binding was saturable and reversible with very low nonspecific binding. [3H]Sulpiride bound to an apparent single population of sites in striatum with a Kd of 3.2 nM and Bmax of 447 fmol/mg protein. Binding sites were localized in the lamina gl...

Calcium and chloride ions stimulated [3H]glutamate binding to quisqualate-sensitive [3H]glutamate binding sites 4-fold, as measured by quantitative autoradiography, whereas 100 mM potassium thiocyanate had no additional effect. In contrast, calcium and chloride had little effect on the binding of [3H](RS)-alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid ([3H]AMPA), but 100 mM thiocyanat...
متن کاملHigh correlation between the localization of [3H]TCP binding and NMDA receptors.
Tritiated N-(1-[2-thienylcyclohexyl)-3,4-piperidine ([3H]TCP), a derivative of the dissociative anesthetic phencyclidine (PCP), has recently been shown to bind specifically to a o-opiate receptor within the central nervous system of the rat (Sircar and Zukin, 1985). Drugs of this class noncompetitively inhibit the excitatory properties of Nmethyl-D-aspartate (NMDA), a glutamate analogue, in the...
